More than a century ago, a French biologist wrote a book called Synthetic Biology, proposing a radical program to mimic the systems of life with non-biological molecules. He made little progress, and his work faded into obscurity. But advances in knowledge since then now make it possible to go far beyond his ambitious dream, not just by mimicking life, but by repairing it, transforming it, and even creating new forms of life. The revolutionary field of synthetic biology already has significant achievements to its credit-in medical treatments, food and agriculture, and materials science. Furthermore, researchers are hard at work on astonishing projects such as
DNA Computers: A single cell may contain the entire code for assembling an organism such as a human. A computer using a DNA "drive" could theoretically store all the world's data in just a few cubic feet of space. Synthetic biology would provide the read-and-write mechanism.
De-extinction: The Jurassic Park scenario could be coming to life-not with dinosaurs, whose DNA is too degraded, but with more recent extinct creatures, like wooly mammoths. The revived species would combine the original with living relatives-for example, elephants.
Xenobiology: DNA happens to be the combination of elements that evolution selected as the carrier for genetic information. There is no reason that other combinations aren't possible, which raises the prospect of all sorts of organisms with novel, even alien, capabilities.
We are living in a golden age of breakthroughs in the life sciences, and nowhere more so than in the field of synthetic biology. In 18 in-depth, half-hour lectures designed for non-scientists, Synthetic Biology: Life's Extraordinary New Worlds explains just what synthetic biology is, what it has done, and what the exciting future may hold. Taught by Professor Milton Muldrow of Wilmington University, the course also considers the profound ethical questions that arise when scientists have the power to create life.
Tools of Synthetic Biology
Ever since the discovery of the double helix structure of DNA in 1953, many of the mysteries of life have slowly yielded to science. The milestones include proof that genetic information flows from DNA to RNA to proteins, in 1958; the combining of DNA fragments from different sources to produce recombinant DNA, in 1972; the first experiments in genetic engineering, in 1973; and the decoding of the human genome, between 1990 and 2003.
These developments, and more, have given researchers the tools to control life at the level of genes, which are the units of DNA that perform specific functions. In this course, you learn that synthetic biology arose from the growing ability to manipulate the machinery of life through techniques such as
CRISPR: Lauded as a world-changing technology, CRISPR is a precise gene-editing tool that won its discoverers the 2020 Nobel Prize in Chemistry. It has revolutionized cancer therapy and other treatments and is being turned to address environmental problems and a host of other diverse uses.
Directed Evolution: Another Nobel prize-winning discovery is directed evolution, a process that greatly accelerates evolution. The idea is to produce proteins with desired traits by letting nature do the work. Nature's path often operates in ways that would be inconceivable to predict.
Metabolic Engineering: By adjusting the metabolism of cells to make them more efficient, valuable products such as insulin can be synthesized in quantity, turning living organisms into cell factories. Today, synthetic biology is supercharging metabolic engineering to deliver a new world of products and medicines.
Dr. Muldrow also covers the use of synthetic biology in textiles, agriculture, and regenerative medicine, and he explains how it greatly accelerated the response to the COVID-19 pandemic in 2020. As soon as Chinese researchers transmitted a digital draft version of the COVID genetic code, scientists around the world had the ability to quickly reproduce it for study. No isolation of a virus from a patient was necessary, nor did researchers have to wait to get a complete sample from a far-off lab. Instead, they could use synthetic biology to create a complete virus genome immediately and set to work developing a vaccine.
The Dangers of Misuse
Powerful tools have a powerful potential for misuse, and synthetic biology is no exception. Professor Muldrow highlights some troubling possible scenarios, including
Embryonic Engineering: Gene therapies allow genetic diseases to be eliminated at their source. But the method also raises the prospect of designer babies with whatever trait is deemed advantageous; creating, for example, super geniuses, athletes, or soldiers. This raises profound social and ethical questions.
Gene Drives: A gene drive is a mechanism to spread a gene or group of genes through a population. This can be done for noble purposes, such as eradicating diseases, but also as part of warfare. Furthermore, a gene drive could unintentionally get out of control, wiping out entire species.
Synthetic Virology: The ability to engineer viruses has enormous potential for benefitting society. But in the wrong hands the technology could be devasting, leading to the revival of scourges such as smallpox. Moreover, the innocent error of a single researcher could have a catastrophic outcome.
Safeguards are already in place to reduce some of these risks. But given the pace of change in synthetic biology and the almost limitless possibilities for harm, much more can be done, as Professor Muldrow discusses.
A Revolution in Progress
Living amid a revolution, it's hard to see where things are heading or how thoroughly society will change. Dr. Muldrow likens today's snowballing advances in synthetic biology to the Cambrian explosion half a billion years ago, when many of the current branches of life abruptly appeared in the fossil record. At the time, the increase in oxygen in the atmosphere due to photosynthesis; the accumulation of molecular and physical adaptations in organisms; the diversity of species relationships; and the appearance of hard body parts in animals, along with other factors, equipped evolution with new variables with which to play. The time was ripe for radical change.

Dogs and cats represent the vast majority of the pets in the United States. While they can behave very differently at times, there's actually a lot of similarities when it comes to training these animals to behave appropriately within a human family. There are just a handful of issues that make up nearly all of the problems that people have with their pet dogs and cats. This means that these problems are very predictable, and fortunately, they're also very preventable. And that's probably the biggest similarity of all. When it comes to behavior problems, whether the pet is a cat or a dog, the easiest, most effective and most efficient approach is prevention. The key is to do as much training as possible, as early as possible. We know what the problems are going to be, we know how to prevent them, and if we prevent the problems, they never form into bad habits. Once a problem has become a habit, it's significantly harder to fix. The treatment is going to use nearly identical methods, it's just going to take much longer than prevention would have. So, what are the problems? Mostly they are behaviors which are natural and necessary for these animals to do, and it's up to us to teach them when, how, and where we would like them to perform these behaviors. Toilet training is the most common issue, especially litterbox training for cats. Destructive chewing and scratching is also common, as are issues of aggression, reactivity, or unwillingness to be handled or restrained. Learn how to prevent all of these problems, and more, using easy and effective methods that are pleasant for you and your pet.

Facilitate puppy play sessions so pups can develop bite inhibition towards other dogs.
Immediately resolve fearfulness and bullying in puppy play sessions.
Utilize Come-Sit-Watch commands to break stares and decrease tension and threatening behaviors between dogs.
Objectively assess the severity of fighting and biting (Fight/Bite Ratio and Bite Scale)
Assess treatment prognosis and the time required to retrain an aggressive dog.
Use the Jolly Routine for classical conditioning.
Organize a Growl Class for reactive (but not dangerous) dogs.

All dogs will display some level of aggression or reactivity at some point in their life, so learning how to address this type of behavior is critical for any dog owner or dog trainer.With a little knowledge and preparation it is easy to prevent almost all aggression, and to quickly address any aggression issues as they begin to emerge, before they develop into problems that can quickly decrease the quality of life for dog and owner alike.These days, I'm pleased to say that many owners understand the importance of early puppyhood socialization. Unfortunately, far fewer owners understand the important of maintaining that socialization through adolescence and adulthood, when dogs naturally become more shy and stand-offish. All too often, a dog will get into a single dog fight that will scare their owners into avoiding other dogs, depriving their own dog of the social interaction they need for rehabilitation. Without ongoing positive social experiences, the dog's sociability only deteriorates further, resulting in self-reinforcing, downward spiral of de-socialization.As soon as you recognize any developing reactivity or aggression you should treat it. The sooner you do, the easier it will be. And the methods used in the treatment of dog aggression are remarkably similar to those used for prevention, except that treatment usually takes dramatically longer.Recently though, new techniques have been developed to accelerate and maximize classical conditioning, progressive desensitization and ultimately, re-socialization. These methods make it possible to rehabilitate dogs as quickly and thoroughly as possible.

'I've learned some of the Photoshop tools, but how do I actually enhance an image, start to finish?'That was the question I saw asked on a Photoshop forum and I thought: 'Good point!' So here is a course designed with just that in mind. I take you through the development of a series of landscape photos, start to finish.We usually start by adjusting a RAW file using the Adobe Camera RAW filter. Then we enhance the photo further in the main Photoshop workspace and go through every step needed to bring it to a final, polished state. Check out the free preview introduction video and see the files we start out with, and then see just how good we can make them by the end.I'll take you through every stage and explain why we are doing things in a particular order. I've given you all the images used on the course to follow along with. I'll show you problems that can crop up and what do about them. Landscape image development brings its own set of problems and opportunities and the various techniques I guide you through take that fact into consideration. This course assumes you have some basic knowledge of Photoshop but I'll also help you there. I've included the notes from 'The Craft of Photoshop: Solid Foundations' which can help to familiarise yourself with the tools we use.As well as giving you a solid grounding in how to develop a landscape photo I'll cover various problems and issues that may arise in your work. We'll also cover things like High Dynamic Range images, stitching panoramic images together (clue - it's got more to do with what you do before you go into Photoshop...), converting images to black and white and how to easily and quickly create a few special effects. Oh, and there's the practice sessions as well...By the end of the course you will have seen and followed along with a series of effective workflows, start to finish. You will gain the confidence to tackle your own images and one day soon you may well be looking at one of your own photos which you've enhanced, and thinking 'That looks fantastic! I did that...'

Also we explore editing parts as an assembly and referencing other components in the assembly
Work with the "Cloud" style file structure is also shown more through out this course while building these projects
Importing files from 3D CAD repositories such as McMaster Carr and Grab CAD Library
If those files cannot be found in those repositories then how to "digitize" them by using reference drawings/documents.
Also using images as canvases in fusion 360 to create some features that are not mentioned in the document that are needed in the assembly

The world of dog husbandry and training is in dire need for another quantum leap! The time is long overdue for all dog professions (breeders, veterinarians, pet store retailers, trainers and shelters/rescues) to utterly reevaluate and refocus their practices and to champion a combined focus on educating prospective and new puppy owners vis a vis puppy raising, so that puppies stay in their original homes instead of being surrendered to shelters as adolescents. Just as we have great owners and not so great owners, it is truly a sobering thought that, by definition, in any profession, when assessed according to any criterion, 50% of professionals fall below median. Dog owners have had an enormous impact on the improved selection of quality products offered in pet stores. As educated consumers, prospective owners can have and probably, will have, similar expectations and effects on breeders, shelters and trainers. From breeders, owners will be looking for puppies that at the very least have been housetrained, chewtoy-trained, manners trained and socialised to people. Similarly, from shelters/rescues, adopters will be searching for dogs that have been housetrained, chewtoy-trained and manners trained during their stay and are now friendlier and more confident. From trainers, puppy and dog owners will be looking for the easiest, quickest, most effective and most enjoyable training techniques that offer immediate and enormous benefits. We really can't keep blaming irresponsible pet owners if no one tells them what to do (before they make predictable and preventable mistakes). The requisite puppy-raising educational materials are readily available but they need to be universally distributed (via email) by all dog professions to all other dog professions and eventually, to all dog owners. Some dog owners might get another puppy. More likely though they will know a friend or neighbor who is searching for one and so, pass along the information. Additionally, put the materials on your company website for free download.The educational materials may be customized with your business name and contact information and so, you may brand your dog business in the community as the one trying to make a difference. Remember, it only takes just one dedicated and persistent person to cause a change. Disseminating information is so doable these days given the technological advances in social media.Dr. Dunbar has created a number of Behaviour Programs for Dog Professionals to evaluate practices and to coordinate and accelerate the distribution of educational materials to prospective and new puppy owners. 1. To encourage Breeders to at least housetrain, chewtoy-train, teach basic manners and socialise their litters and especially, to preferentially breed for longevity. Longevity is the best indicator of physical and behavioural health. It is simply not right that so many pure-bred dogs predictably die so young. "A live dog is better than a dead lion." Ecclesiastes 92. To encourage Veterinary Practitioners to take full advantage of their most unique and enviable position of seeing every breeder, most prospective puppy owners (existing clients) and nearly every new puppy owner on several occasions when puppies are brought in for immunization shots. This is the optimal time for puppies and their owners to receive an educational vaccine. Of all the dog professions, veterinarians have by far the best opportunity to prevent the development of utterly predictable and easily preventable dog behavior and temperament problems. "It is easier to build strong puppies than to repair broken dogs." (With apologies to Frederick Douglas.)3. To encourage Pet Store Retailers to disseminate the same educational materials (i.e., instructions for use of dog products) to prospective or new puppy owners that come to purchase food and supplies. For example, puppy owners need to know why they are buying a dog crate - to prevent mistakes around the house, to autoshape chewtoy chewing and to predict when the puppy needs "to go" so that it can be taken to a toilet area and trained to eliminate on cue.4. To encourage all Shelters/Rescues to adopt an in-house, behavior and training programme, such as Open Paw - at the very least including, housetraining, chewtoy-training, basic manners and ongoing socialization with people and other dogs. Simply housing dogs in cages is not sufficient. For example, without regular toilet breaks, a previously housetrained dog will be forced to soil its living area. Sheltering is supposed to make homeless dogs more adoptable, not less adoptable. But even more important, shelters/rescues must strive to stop the problem at the cause and so, reduce shelter input. Of all the dog professions, with their enormous mailing lists, animal charities have the best opportunity to disseminate puppy-raising materials to dog people.5. To encourage Dog Trainers to be the people that catalyse the change in their communities by being more proactive in promoting their businesses and at the same time, educating prospective and new puppy owners by emailing the educational materials to all clients, all other dog professionals, all local media and to anyone and everyone.

Instead of Gamsol, we will use a burnisher to help lay down the tooth of the paper and produce a more unified look.Section 9, Drawing a Landscape - Draw an interesting landscape scene and learn how to produce effective, subtle gradients as well as layering and blending methods.Section 10, Drawing a Portrait - Layer up colors to produce a realistic and beautiful portrait drawing. This section begins with exercises to learn how to produce different skin tones.Besides about 25 hours of helpful video instruction and demonstration, this course also includes all the photo reference support files and a booklet PDF complete with worksheets to follow along with, including mark making worksheets, art project worksheets, and color theory worksheets.
